Yes, the A52Codec is necessary.
Perian's supported formats web page shows AC-3 as playable but not decodable, then recommends A52Codec, and their development tracking system has an open ticket created by one of the project's developers for AC-3 support without A52Codec.
Just double-checked with one of our AC-3 test files - it comes up without audio when A52Codec is not installed but works great when A52Codec is installed.
From the looks of it, AC-3 may be fully supported by a future version of Perian. The underlying ffmpeg source code can be built to support it, it's the hooks between that source and QuickTime that the Perian team has to work on.
